Study reveals African men at higher risk for prostate cancer
A groundbreaking study has uncovered genetic risk factors that contribute to the high prevalence of prostate cancer among African men.
The research, the largest of its kind, recruited 7,500 men from eastern, southern, and western Africa, aiming to explore why men of African descent face a significantly higher risk of developing the disease.
Understanding the Genetic Risk
Prostate cancer is disproportionately common among men of African descent, with studies from both the United States and various African nations showing higher rates of the disease. Men of African and African Caribbean ancestry not only face an increased risk of prostate cancer but also are more likely to develop aggressive forms of the disease at a younger age compared to their European counterparts. In Africa, prostate cancer accounts for nearly one-quarter of all cancer cases.
The research, led by medical scientist Wenlong Carl Chen, sought to identify the genetic factors that underlie this increased susceptibility in African men. Specifically, the study aimed to understand the genetic makeup that contributes to higher prostate cancer risk in this demographic. It focused on both hereditary risk factors, such as rare genetic mutations, and more common genetic variants known as single nucleotide polymorphisms (SNPs), which are associated with increased disease risk.

“Despite the high burden of disease in Africa, genetic studies of prostate cancer in continental African populations have been limited,” said Chen. “This study is crucial in filling this gap, as the technology required for such studies is expensive and often not available in many African countries.”
A Unique Approach to Genetic Research
What sets this study apart is its focus on African populations, which exhibit much higher genetic diversity compared to other groups. As the birthplace of humanity, Africa’s population has had more time to accumulate a wide range of genetic variations, making it essential to include African populations in genetic research for a deeper understanding of the disease.
The research was part of a collaborative project called the Men of African Descent, Carcinoma of the Prostate (MADCaP) consortium, established in 2016. The consortium brought together hospitals and universities from five African countries—Nigeria, Ghana, Senegal, Uganda, and South Africa—as well as US-based institutions, with funding from the US National Institutes of Health (NIH). The study included 3,963 prostate cancer cases and 3,509 controls, with bio-samples and genetic data collected across these regions.
Key Findings
The study identified three specific genetic loci (regions of the genome) strongly linked to increased prostate cancer risk in African men. While these loci had been previously identified in other populations, the genetic signals found in this research were unique to African populations and would not have been discovered without focusing on these groups.
Additionally, the study found that the genetic factors influencing prostate cancer risk vary across different African regions, highlighting the importance of geographical diversity in genetic research.
Implications for Future Treatment and Screening
The findings have significant implications for the future of prostate cancer research and treatment. According to Chen, the study paves the way for the development of genetic tools that could help assess an individual’s risk of developing prostate cancer. These tools could lead to earlier detection and more personalized treatment options for those at high genetic risk.
For example, similar genetic screening methods have already been used in breast cancer research, where women with higher genetic risks are eligible for early screening programs. Chen believes that similar strategies could be implemented in Africa to combat prostate cancer.
“Our findings highlight the importance of studying diseases in populations with the highest disease burden,” Chen added. “By identifying genetic risk factors unique to African populations, we are laying the groundwork for the development of diagnostic and preventative tools tailored to the needs of these populations.”
As this study progresses, it is expected to inspire more research into the genetic underpinnings of prostate cancer, ultimately leading to better prevention, detection, and treatment strategies for African men, who face an urgent and growing health challenge.
